Research Highlights

 

Oil Palm DNA chip
 

 

>

Four genetic linkage maps of oil palm have been constructed.

>

A total of 450,000 oil palm sequences have been generated. 

>

About 4,000 of these clones with known identities were printed on the MPOB DNA chip. The MPOB DNA chip has been used to profile gene expression in oil palm tissues, including leaf, inflorescence and callus. 

>

In cytogenetics, the stages of pollen mother cells (PMC) or microsporocytes suitable for anther culture to produce haploid plants were successfully identified. 


Core Facilities & Services

>

Genome Analysis Laboratory for Oil Palm (GALOP)

This facility will be set up as a resource centre to serve both the Industry and MPOB. The following facilities will be housed in GALOP:

- DNA Banking - to deposit reference materials as cultures, DNA or tissues for future reference
- DNA Fingerprinting - use of DNA probes as quality control tools in oil palm tissue culture and breeding
- Chromosome Painting - a technique to assist breeders in hybrid breeding programmes.
- DNA Sequencing

The core facility operates three ABI377 automated sequencers (with 36-96 samples throughput) which produce sequence data that is returned to the investigator as a sequence file (.seq file format or ABI format).

At present the sequencing service is only offered to researchers at MPOB.

DNA sequencing is carried out on plasmids and phage stocks.
Sequencing reaction run is based on dye-labeled terminator chemistry.
Reads up to 900 bases obtainable per reaction.

Cytogenetical Analysis

  • Chromosome Painting In Oil Palm Hybrids
    Genomic in situ hybridization (GISH) method was developed to distinguish between oleifera and guineensis chromosomes. Hence this is a useful method for oil palm breeders to assess the genome composition of backcrosses and oil palm hybrids.
  • Flow Cytometry Applications
    Flow cytometry is a rapid and convenient technique that allows accurate determination of (1) nuclear DNA content and genome size in plants and (2) the ploidy level of any organisms. It is a useful tool for research ranging from evolutionary studies to genome mapping and ploidy manipulation.
  • Plant Mitosis and Meiosis: Cytogenetical Analysis
    Cytogenetics is a branch of genetics involved with the study of chromosomes and cell division. Fluorescent and non-fluorescent dyes can be used to visualize the chromosomes. Mitosis and meiosis are both processes of cell division and occur in somatic and reproductive cells respectively. By doing cytogenetical analysis, mitotic and meiotic cells can be analyzed to observe any abnormalities in chromosomes morphology or process.
